The Vignali Lab primarily studies inhibitory mechanisms in cancer using mouse models and occasionally patient samples and datasets (pre-/post-treatment with immunotherapies), with a focus on inhibitory receptors (LAG3 and PD1) and regulatory T cells (Tregs). We also study mechanisms of T cell exhaustion. We make heavy use of sophisticated mutant mouse models, systems immunology, transcriptomic analysis (scRNAseq), super-res microscopy and multispectral imaging and transcriptomics. We are also focused on the development of novel immunotherapies, and engineer our own antibody-based therapeutics.
